Gene discovery reveals importance of eating your greens

Mar. 3, 2013 ? Eating your greens may be even more important that previously thought, with the discovery that an immune cell population essential for intestinal health could be controlled by leafy greens in your diet.

The immune cells, named innate lymphoid cells (ILCs), are found in the lining of the digestive system and protect the body from 'bad' bacteria in the intestine. They are also believed to play an important role in controlling food allergies, inflammatory diseases and obesity, and may even prevent the development of bowel cancers.

Dr Gabrielle Belz, Ms Lucie Rankin, Dr Joanna Groom and colleagues from the Walter and Eliza Hall Institute's Molecular Immunology division have discovered the gene T-bet is essential for producing a population of these critical immune cells and that the gene responds to signals in the food we eat.

Dr Belz said the research team revealed T-bet was essential for generating a subset of ILCs which is a newly discovered cell type that protects the body against infections entering through the digestive system. "In this study, we discovered that T-bet is the key gene that instructs precursor cells to develop into ILCs, which it does in response to signals in the food we eat and to bacteria in the gut," Dr Belz said. "ILCs are essential for immune surveillance of the digestive system and this is the first time that we have identified a gene responsible for the production of ILCs."

The research was published today in the journal Nature Immunology.

Dr Belz said that the proteins in green leafy (cruciferous) vegetables are known to interact with a cell surface receptor that switches on T-bet, and might play a role in producing these critical immune cells. "Proteins in these leafy greens could be part of the same signalling pathway that is used by T-bet to produce ILCs," Dr Belz said. "We are very interested in looking at how the products of these vegetables are able to talk to T-bet to make ILCs, which will give us more insight into how the food we eat influences our immune system and gut bacteria."

ILCs are essential for maintaining the delicate balance between tolerance, immunity and inflammation. Ms Rankin said the discovery had given the research team further insight into external factors responsible for ILC activation. "Until recently, it has been difficult to isolate or produce ILCs," Ms Rankin said. "So we are very excited about the prospect for future research on these cells which are still poorly understood."

ILCs produce a hormone called interleukin-22 (IL-22), which can protect the body from invading bacteria, Dr Belz said. "Our research shows that, without the gene T-bet, the body is more susceptible to bacterial infections that enter through the digestive system. This suggests that boosting ILCs in the gut may aid in the treatment of these bacterial infections," she said.

ILCs help to maintain a 'healthy' environment in the intestine by promoting good bacteria and healing small wounds and abrasions that are common in the tissues of the gut. They may also have a role in resolving cancerous lesions. "The discovery of these immune cells has thrown open a completely new way of looking at gut biology," Dr Belz said. "We are just starting to understand how important these immune cells are in regulating allergy and inflammation, and the implications for bowel cancer and other gastrointestinal disorders such as Crohn's disease," she said.

"Understanding the biology of ILCs and the genes that are essential for generating them will help us to develop methods of targeting these cells," Dr Belz said. "This might include boosting ILCs in situations where they may not be active enough, such as infections or some cancers, or depleting them in situations where they are overactive, such as chronic inflammatory disease," she said.

Russia to put dead whistleblower on trial

MOSCOW (Reuters) - A Russian court will start the posthumous trial of a dead anti-corruption lawyer next week after ignoring calls by his family and lawyers to abandon a case they say is absurd and politically motivated.

Sergei Magnitsky's death in custody in 2009, after he had complained repeatedly of being denied medical treatment, has damaged Russia's image and strained ties with the United States.

But Moscow's Tverskoy Court said after a pre-trial hearing on Monday that the trial itself would open on March 11, a court spokeswoman said.

Lawyers say Magnitsky, who was 37 and was accused of tax fraud after investigating similar claims against his accusers, will be the first dead person to go on trial in Russia.

"The trial is indeed absurd," said lawyer Alexander Molokhov after the court rejected his application to defend Magnitsky.

The court had already appointed a legal team to defend Magnitsky after his own lawyers refused to take part in a trial which his relatives say is politically motivated.

Magnitsky's mother, Natalya, has said previously that the case is a farce and her lawyer Nikolai Gorokhov likened the proceedings to "dancing on the grave of a dead man".

The allegations that Magnitsky, who was a lawyer at Hermitage Capital Management investment fund, had committed tax fraud were made shortly after he accused state officials of a $230 million theft by setting up bogus tax refunds.

Critics say the trial is meant to discredit Magnitsky as well as Hermitage owner William Browder, who will be tried in absentia.

President Vladimir Putin has said that Magnitsky, who was 37, died of heart failure but his presidential human rights council has said the lawyer was probably beaten to death. No one has been convicted over Magnitsky's death.

His death has also weighed on ties between Moscow and Washington. U.S. lawmakers passed a bill last year that attempts to punish Russians who were involved in his case and are accused of violating human rights.

Russia, in turn, passed similar measures aimed at punishing Americans suspected of violating human rights.

It also banned U.S. families from adopting Russian children after the death this year of Russian-born Max Shatto, who was adopted by a U.S. family in Texas.

The case against Magnitsky was initially closed after his death in November 2009, but authorities reopened it in 2011 as international criticism over his death - and Russia's apparent reluctance to hold anyone criminally responsible - mounted.

Magnitsky and Browder were charged last year, weeks before the United States adopted the so-called Magnitsky Act which also imposes asset freezes and bars from entry to the United States anyone suspected of a role in his death.

Human-made material pushes the bounds of superconductivity

Mar. 3, 2013 ? A multi-university team of researchers has artificially engineered a unique multilayer material that could lead to breakthroughs in both superconductivity research and in real-world applications.

The researchers can tailor the material, which seamlessly alternates between metal and oxide layers, to achieve extraordinary superconducting properties -- in particular, the ability to transport much more electrical current than non-engineered materials.

The team includes experts from the University of Wisconsin-Madison, Florida State University and the University of Michigan. Led by Chang-Beom Eom, the Harvey D. Spangler Distinguished Professor of materials science and engineering and physics at UW-Madison, the group described its breakthrough March 3, 2013, in the advance online edition of the journal Nature Materials.

Superconductors, which presently operate only under extremely cold conditions, transport energy very efficiently. With the ability to transport large electrical currents and produce high magnetic fields, they power such existing technologies as magnetic resonance imaging and Maglev trains, among others. They hold great potential for emerging applications in electronic devices, transportation, and power transmission, generation and storage.

Carefully layered superconducting materials are increasingly important in highly sophisticated applications. For example, a superconducting quantum interference device, or SQUID, used to measure subtle magnetic fields in magnetoencephalography scans of the brain, is based on a three-layer material.

However, one challenge in the quest to understand and leverage superconductivity is developing materials that work at room temperature. Currently, even unconventional high-temperature superconductors operate below -369 degrees Fahrenheit.

An unconventional high-temperature superconductor, the researchers' iron-based "pnictide" material is promising in part because its effective operating temperature is higher than that of conventional superconducting materials such as niobium, lead or mercury.

The research team engineered and measured the properties of superlattices of pnictide superconductors. A superlattice is the complex, regularly repeating geometric arrangement of atoms -- its crystal structure -- in layers of two or more materials. Pnictide superconductors include compounds made from any of five elements in the nitrogen family of the periodic table.

The researchers' new material is composed of 24 layers that alternate between the pnictide superconductor and a layer of the oxide strontium titanate. Creating such systems is difficult, especially when the arrangement of atoms, and chemical compatibility, of each material is very different.

Yet, layer after layer, the researchers maintained an atomically sharp interface -- the region where materials meet. Each atom in each layer is precisely placed, spaced and arranged in a regularly repeating crystal structure.

The new material also has improved current-carrying capabilities. As they grew the superlattice, the researchers also added a tiny bit of oxygen to intentionally insert defects every few nanometers in the material. These defects act as pinning centers to immobilize tiny magnetic vortices that, as they grow in strength in large magnetic fields, can limit current flow through the superconductor. "If the vortices move around freely, the energy dissipates, and the superconductor is no longer lossless," says Eom. "We have engineered both vertical and planar pinning centers, because vortices created by magnetic fields can be in many different orientations."

Eom sees possibilities for researchers to expand upon his team's success in engineering human-made superconducting structures. "There's a need to engineer superlattices for understanding fundamental superconductivity, for potential use in high-field and electronic devices, and to achieve extraordinary properties in the system," says Eom. "And, there is indication that interfaces can be a new area of discovery in high-temperature superconductors. This material offers those possibilities."

Funding from the U.S. Department of Energy Office of Basic Energy Sciences, National Science Foundation, and the Air Force Office of Scientific Research supported the researchers' work. Eom's collaborators include Eric Hellstrom's and David Larbalestier's group at Florida State University; and Xiaoqing Pan's group at the University of Michigan.

US seizes 2,200 pieces of artwork in NJ

NEWARK, N.J. (AP) ? The massive trove of artwork arrived in New Jersey last year from Texas on an 18-wheeler ? its contents a mystery.

After five days of inventory, the haul proved to be significant: more than 1,100 pieces of art, mostly works by some of the nation's most influential photographers, including Alfred Stieglitz and Edward Weston.

The valuable shipment was moved again to New York, but instead of being carefully exhibited in a home in Spain as planned, it ended up in the hands of federal authorities. By the end of an investigation, authorities had seized more than 2,200 works of art appraised at nearly $16 million.

On Friday, the U.S. Attorney's Office in Newark announced a Feb. 22 court filing that alleges the artwork was purchased with money collected in a scheme that sold fake credits for renewable energy. The complaint seeks to force the owner to forfeit the works.

Authorities said they were seized from a warehouse in New York in July weeks after being moved from Newark and readied for shipment to Spain via Amsterdam. The works include numerous prints by Stieglitz, including one of his famous artist wife, Georgia O'Keefe, that sold for $675,000. An Edward Steichen print titled "Greta Garbo for Vanity Fair Hollywood" was purchased for $75,000.

Authorities allege the artworks' owner, Philip Rivkin, used money fraudulently funneled through his Houston-based company, Green Diesel, to buy the art. Rivkin has not been formally charged with a crime.

Kyle Sampson, a lawyer for Rivkin, said Friday he was aware the forfeiture action was filed.

"We have not received formal notice of the forfeiture action, but I am aware that a complaint has been filed," he told The Associated Press. "We've been working for some time with the EPA in an effort to resolve their concerns and any other outstanding issues there may be."

A lawyer for Green Diesel declined comment in an email message.

Rivkin was owner and CEO of Green Diesel and a company called Fuel Streamers. Authorities allege in the complaint that Rivkin and others claimed the companies produced, purchased and imported renewable fuel. But authorities say the fuel did not exist.

By federal standards, the national stock of gasoline and diesel must contain a certain percentage of renewable fuel. Authorities allege oil companies, including Shell and Exxon, have reported a total of $78 million in losses from buying fake renewables from Green Diesel.

According to the complaint, an unnamed assistant for Rivkin told authorities that Rivkin controlled all financial accounts. "Money would come in and he (Rivkin) would move it out," the assistant told authorities.

Authorities allege Rivkin used Green Diesel's bank account and other accounts to buy at least $18 million in artwork. Rivkin tended to buy his photographs in groups, making it difficult to trace individual purchases, but authorities said they have matched 1,590 pieces of art to one credit card and four bank accounts.

In April 2012, the federal Environmental Protection Agency issued a notice of violation to Green Diesel, alleging it improperly generated credits that track and boost renewable fuel production. Authorities claim Rivkin arranged to have the art shipped out the year before, after learning of the investigation.

An EPA spokesman did not return a call for comment.

According to the complaint, Rivkin's assistant said Rivkin started purchasing vintage photographs after receiving the "EPA money." Much of the artwork was shipped from Houston to Newark and eventually bound for Spain, authorities said.

The assistant traveled to Geneva and Barcelona with Rivkin, the assistant told authorities. Rivkin stayed in Spain and when the assistant returned to the U.S. noticed that Rivkin's office was cleaned out and numerous documents were shredded, authorities said.
